No it doesn't. It allocates space for the directory tables when
formatted, one of the reasons for the fast directory handling. But then
it doesn't use any more space for this when actually writing the files.
It may appear to give you less usable space on a freshly formatted
partition, but in fact you usually get slightly more.

Any unused directory space is freed up if the partition becomes full.
