FIGURE 1-3. (A) Restriction enzyme recognition sites for seven
different restriction enzymes are shown. The enzymes cleave the DNA
strands where indicated by the arrows. DNA fragments with
complementary ends produced by cutting with the same restriction
enzyme can be ligated together thus allowing the construction of
recombinant DNA molecules. (B) DNA fragments in 1 microgram of
purified polyoma virus DNA digested with EcoRI (lane 1) or HindIII
(lane 2) and 10 micrograms of human placental DNA digested with
HindIII (lane 3) were size-fractionated by agarose gel
electrophoresis. The resolved DNA fragments were visualized by
illumination with UV light after staining with ethidium bromide.
Comparison of the band positions to the positions of DNA markers of
known size running in the left-most lane allows determination of
fragment size.
